The Defense Mastery Tree (Season Three) is one of the three Mastery branches and the current iteration of the defensive mastery branch. They were introduced with the V1.0.0.152 update of Preseason 3 on December 4th, 2012.

It would take 37 points to max out the Defense Tree.
