Masteries, like Runes, are enhancements that summoners provide their champions. They are divided into three trees (groups): Offense, Defense, and Utility. Each group has six tiers. Within a group, all but the first tier are locked when you first start, with a successive tier opening with each 4 points spent in the same group.

A summoner can put one point per summoner level (up to level 30) into masteries. Due to the number of masteries available, this means that a summoner cannot have access to all of the masteries at once, nor even an entire group all at once.
